looks like a skin infection. I supect its fungal but cant be too sure.

I would get some multi-cure as it can deal with both fungal and bacterial issues. If you look a bit closer an the wound seem 'fluffy' then its fungus and go for some fungus killer. if not then go for some anti-bacterial stuff. if you cant decide then you go with the multicure

the tank originally had 4 l333 about 4cms a 5cm pepp heaps of guppies and a pair of pepp
but now for no reason over a few days they all died except for male pepp and one guppy they look as though they are not affected at all.
i check water parameters all good

ammonia 0 -.25
nitrite 0
nitrate 0
is there any reasons at all that this would happen?

also they are livin in a standard 3 ft with aqua one cf 1000 filter

Your getting a reading for ammonia, this could be a possible cause, any reading above 0 is toxic to fish. The nitrate reading is 0 I would expext some reading for nitrate in an established tank. Your tank may have gone into a cycle.
